Title: Where did my Bitcoins go?! HELP!!!

I'm new to this and downloaded Bitcoin.  I sent 1 bitcoin to myself from a website to make sure I was doing it correctly and that it worked.  No problem I got the 1 coin.  Then I sent a large amount and NOTHING.  The website that sent it says it's sent but I'm not seeing it in my wallet.  WTF?!?!

Is your client up-to-date downloading the block chain?

Do you see the transfer in blockexplorer?
Thank you for your reply.  I did a search and it says there was no transactions for that address.

The next thing to check is the list of pending transactions. You can find that here:
http://bitcoincharts.com/bitcoin/
If it's not in the list of pending transactions and it's not in the public hash chain, then most likely the other end has not sent the bitcoins.

A transaction should appear in the list of pending transactions with a few minutes, unless a lower than standard transaction fee was paid. In that case, it can take as long as a day to get into the list of pending transaction. Then it takes typically about ten minutes to get in the public hash chain, but it can take longer if there are lots of higher priority transactions or the network just has bad luck.

Ok thanks for your help.  I assumed it would always be a quick as the first transaction.  When it didn't appear I got a little worried.  I'll just be patient because everything seems to be fine.
