В случае SARS-CoV-2 проникновение вируса осуществляется двойным образом с помощью белка-шипа (далее шип) внутри вирусной мембраны и ангиотензин-превращающего фермента 2 (ACE2) в мембране клетки человека.  И шип, и ACE2 известны как гликопротеины - белки, содержащие длинные молекулы, называемые  гликанами. Влияние гликанов на биомолекулярные пути многочисленно, и мы до сих пор не до конца понимаем каждый процесс, в котором они участвуют. Однако было показано, что при SARS-CoV-2 они играют важную роль в функционировании шипа[1].

Часть шипа, которая связывается с его рецептором, ACE2, называется связывающийся с рецептором домен (RBD).  Чтобы смягчить инфекцию SARS-CoV-2, многие искусственно созданные антитела конкурируют с ACE2 за связывание RBD, напрямую блокируя событие связывания ACE2: RBD, необходимое для проникновения вируса.

В то время как структурные данные, представленные Пинто и др раскрывает некоторые ключевые идеи о том , как S309 взаимодействует с RBD, он предоставляет только статический снимок комплекса RBD:S309. Лаборатория Chodera проводит моделирование молекулярной динамики (MD) (с использованием OpenMM) для изучения движения белков во времени. Однако моделирование MD является дорогостоящим с вычислительной точки зрения, поэтому, если мы хотим изучать движение белка в течение длительного времени, нам нужно много вычислительных ресурсов. Вот тут-то и появляется Folding@home!

Благодаря огромной совокупной вычислительной мощности Folding@home мы можем генерировать данные моделирования в беспрецедентных масштабах.  Затем мы можем использовать данные моделирования, чтобы сделать подробные видеоролики о белках в движении и, что более важно, получить представление о конформационных состояниях, которые может принимать интересующий белок. Почему нас волнуют конформационные состояния белка?  Каждый белок принимает ряд конформаций, где каждая конформация диктует взаимодействия белка с его партнерами по связыванию, тем самым влияя на функцию белка.  Более того, диапазон конформационных состояний может изменяться в присутствии нового партнера по связыванию.

Мы можем применить эти идеи к RBD и попытаться понять механизм действия S309 с точки зрения изменения конформации белка.  Мы предполагаем, что когда S309 связывается с RBD, он изменяет набор конформационных состояний RBD, так что RBD больше не связывается со всеми теми же партнерами, нарушая способность вируса проникать в клетки человека. Чтобы исследовать эту гипотезу, мы запустим моделирование RBD (проект 17307), ACE2 (проект 17308), RBD: ACE2 (проект 17309) и RBD: S309 (подготовка к F@h в процессе разработки) и сравним диапазон конформационных состояний для RBD в этих симуляциях.

В результате каждого моделирования будет выведен набор всех конформаций, в которых конструкция находилась во время моделирования. Каждая конформация может соответствовать движению одного атома внутри структуры белка. Учитывая, что белки могут состоять из тысяч атомов, объем данных результатов моделирования будут чрезвычайно большим. Чтобы разобраться в этих огромных объемах данных, мы создаем модели состояния Маркова (МСМ), которые представляют собой основы для анализа динамических систем, таких как белки.  МСМ позволяют группировать похожие конформации как «состояния».  Как только данные организованы таким образом, мы можем определить, как конформационные состояния RBD изменяются при связывании S309 и помогают ли эти наблюдения объяснить механизм действия S309.

Until now, the exchange has allowed American users to access its platform. A US resident just had to click the "I'm not [American]" button to create an account on Binance.com.

Binance currently sends emails to US residents based on their IP addresses.

"We noticed that your account may be linked to an IP address from the United States. In accordance with regulatory requirements we are unable to provide services to citizens or residents of the USA […] If you are a us citizen or resident, withdraw your assets from your account within 90 days. We recommend that you consider using Binance U.S. or other platforms in the United States, " the letter says.

Binance took these steps after the US government launched two lawsuits against the BitMEX crypto derivatives exchange.
