I have a rig with 6x7950 Sapphire cards connected to an MSI GD65 motherboards through powered risers.

Do I need to use 3 connections per card, i.e 2 6pin connectors and the molex connector in the riser?

It can be, but I don't have it in my area

Do you think the Gigabyte GA-Z77X-UP4 TH would be able to support 6 GPU's
