I moved some bitcoins from my mtgox account to a desktop Multibit wallet, then exported the keys and stored them on a hard drive. If I add more bitcoins to this wallet, do I have to delete the previously exported file and export a new one?

That depends on whether you generate new addresses for the new transactions, or re-use the addresses that you already backed up.

let's say I don't make a new address. Just add more bitcoins to an existing wallet.

the backup is actually like a proof of ownership of your wallet.

the wallet and it's balance are maintained through the bitcoin network... with the "private key" you can at anytime claim that wallet to be yours and debit coins from it.

so answer is: NO, you don't need to make new backups
