My motherboard has 2 PCI E X16 Slots about 2 years ago. According to my vendor, he said that if i put in two Graphic cards, One will run at X16 and the other will run at x8.

Will this affect my mining speed.

Im planning to buy a new rig with the below motherboard and plan to put 4 Graphic cards on it. Is this suitable if i put 4 HD 5870.

You can run gpu's for mining at x1 if you want.

It would only affect your gaming performance, mining doesn't care about x1, x8, x16.

If you look around at the people who use extenders to keep their GPU's cool, most of them are only using the PCI-e x1 ribbons. It doesn't require much bandwidth to the card to mine.
