Make the Bitcoin, don't let the Bitcoin make you.

I have used bitinstant with success - although their website is being upgraded right now. I bought $2000 per day by depositing cash at WalMart via Moneygram. I had the BTC before I even got out of the store.

The website is terrible btw and didn't inspire confidence - hopefully the new one will be more user friendly and professional. It's the fastest way to buy BTC in reasonable amounts (I got 24 a day @~75USD)

Second thebrit's BitInstant suggestion. Some of their customers have been having trouble lately, though. I have a walkthrough for buying on BitInstant: http://www.bit-buddy.com/buying-bitcoins-with-bitinstant.html.

If you have a checking or savings account you're willing to link, Coinbase is pretty user-friendly: https://coinbase.com/?r=51a5b0d3867ba8db8e00000c. Linking your account there is very much like setting up on PayPal. I have a walkthrough for their service as well: http://www.bit-buddy.com/buying-bitcoins-on-coinbase.html

+1 on coinbase.com

You can link it to your bank account via ACH. I've been using them for a while now.

What kinds of transaction costs does each option have? Coinbase is around 1% and BTC/USD exchanges are around .5%, for comparison.
