Can you lock your computer, and have your miners still running full speed?

Yes. Alot of people Remote Desktop to their mining rigs. This means as soon as they close the RDP connection, it essentially locks the computer.

Not if you use a graphical screensaver - it will slow down the mining. Best is a plain black screen.
