I've created a USB-stick with tails and Electrum on it. Then I created a cold storage wallet in it to be only used offline.

I've read a lot on this forum, but I'm confused about this:

What do I need to back up in case the stick breaks or gets lost?

I've written down the 12-word seed on paper, is that enough?

If you didn't enable persistence, you have to backup all the files that you have generated if you want to save them.

The seed can restore all your addresses. Back it up and store it in a safe place and you will be alright.

Yes the seed is all you need http://docs.electrum.org/en/latest/faq.html

So just to be sure, if I ever lose my USB-Stick, I just install Electrum on my computer and restore a wallet with the seed and I'll have access to all my BTCs. Right?

I have enabled persistence in Tails, does that mean that I have to back up something else additionally?

Yes but the labels will not be restored. To restore those too save a copy of the wallet file via file menu > save copy. Alternatively enable the labels plugin so that your encrypted labels are uploaded to a third party server:

If you have a large amount of btcs then i would recommend you to make 2-3 separate usb backups, you can't trust a single usb, they can fail anytime or get corrupt if they are just sitting there. So 2 paper backups of the seed and 2-3 usb backups of the seed and the wallet.dat file. You just have to worry about the seed file that's all.
