I'm certainly no expert, but from my understanding there's no definite way of knowing. You could probably do some analysis on the blockchain to determine what other addresses are owned by the same person and get an estimate from that. It would be a lot of guesswork though.

You can only know the coins in that address. To know the coins in a wallet.dat you must first know all the addresses of that wallet! Sure, you can guess somethings, by looking at transactions and etcetc, but only guesses, and maybe totally wrong.

If a subsequent transaction spending those funds also includes other inputs, you might be able to conclude that the recipient also has the funds from the other inputs as well.  That is not guaranteed though as if the wallet is a shared (hosted) EWallet, it will combine inputs which may be funds received from numerous unrelated transactions.
