I am currently mining 50btc (and possibly mtred, if it get back up) with a Radeon HD 5850 and getting about 350MH/s. What difficulty should I be using on my miners?

Thank you for asking this, I'm aswell registered in 50btc and I since the begining i'm on difficulty 2. tonight I will set the difficulty to 1 and monitor my result with the chart in the dashboard to see what works better for me. According to the FAQ if you have only one 5850 you should stick to difficulty 1.
