I'm thinking in buying 6 RX 480 and a MSI Z170a SLI LGA 1151 MOBO? Will I have any hardware compatibility issue to connect 6 cards?

Yeah, it should work just fine. Try to find some tutorials that show how to set up this MOBO's BIOS for multi GPU usage and mine away !
Though, there are cheaper options for mining motherboard (for hitting the RoI faster).
