Hey,

  Can I run these cards in one machine on one instance of CGminer?  Or would I have to run 2 instances of CGminer?

Yup, just be sure to specify different clock speeds / thread counts in your launch .BAT file. The cards can be picky... my XFX 290's only like 947 core 1250 mem, while my 280x's will do 1125 and 1500 easy
