So, I've just transferred about 65 btc to my silk road account. I have my cart ready, but I'd like to know a bit more about it before I buy, as it's my first transaction on silk road.

1. How do I put the coins in escrow?
2. The seller I am buying from requires people who have made fewer than five purchases to finalize early. I am aware that many scammers ask for that, but I have researched this vendor and found nothing but positive feedback, so I'm confident they're not  scammer. How would one finalize early?
3. As you have probably figured out, I haven't really purchased anything on SR yet. I'm afraid to go ahead and buy it because I'm worried I'll make a mistake and lose all my bitcoins. How easy is sr's checkout process, in your experience? How would it differ from a site like Amazon?

I don't have a clue as I've never completed a purchase on Silk Road.  However, you'd probably be better off asking on the Silk Road forums.

1. Escrow is automatic.  The coins are automatically placed in escrow when you check out.
2. After you submit your order, a "Finalize" link will appear.  Click that if you want/need to finalize early.
3. The checkout process works like any other shopping cart.

You won't be able to finalize your order until the seller accepts your order, then the finalize option will show up when you click on the "my orders" link.

Well, you're in the newbie forum, so they're not going to have a lot of posts. And also, think - could the advice they're giving you potentially lose you bitcoins?

Never sign for a package from the SR. If the delivery man asks you to sign he is either making a mistake or you are experiencing a controlled delivery.
