Do you need any confirmations of your Bitcoin transaction to validate your bet and receive the payout ?
Depends on your transaction fee, LB's bet engine will decide for it, for example if that engine detects that your transaction fee is not standard so for sure it will wait for 1 confirmation then you will see your bet(s) are coming down else you will see your bets instantly. Edit: LB sends back payouts instantly....sometimes it might takes seconds because payback engine could be busy with processing a lot of paybacks, but if you didn't see your bets for minutes... for example for more than 3-4 minutes you can get help by moderators and if moderators were not on the line, you can drop a quick text at support@luckyb.it, then they will handle it.

Just curious, Is that possible if you lowering min bet ? Let say min bet BTC 0.0005
What will be a direct impact because of that ?
The smaller the bet, the larger % the transaction fee will be. You need to pay 0.0001 bitcoin for your bet, and another 0.0001 will be subtracted from your winning.

What will be a direct impact because of that ?

lowering amount to 0.0005 btc means that you sucked up coz when you bet 0.0001(fee) cut out and when lb pays again the fee`s paid so you wont win anything unless you got big multiplier`x

I think every site have to confirm every transaction. Not all sites. Some let you play without any confirmations but require a confirmation to do a withdraw...

How exactly will that work? Wouldn't there be a risk of people double spending their coins, and take advantage of that? Most offchain sites require your deposits to have confirmations before you can withdraw and will invalidate bets made against doublespent funds. Lucky Bit will surely take the appropriate precautions.
