Barcelona will not overtake Real Madrid at the top of the standings even if they win in the next El Clasico, but maybe that will become a reality if Real Madrid loses 3 consecutive matches while Barcelona takes every point available in every match for the rest of the season. Such a scenario is very difficult to expect, I think it is almost impossible because Real Madrid will do everything to stay at the top of the standings.

Real Madrid will not be under big pressure in El Clasico even though they could lose against Barcelona.
